Vanilloid Receptor -Independent Induction of Cancer Cell Apoptosis by Capsaicin

Abstract
Effect of capsaicin, a vanilloid receptor (VR) agonist, on apoptotic cell death was investigated in HepG2 human hepatoblastoma cells. Capsaicin induced apoptosis in a dose-dependent manner evaluated by flow cytometric assay of hypodiploid nuclei stained with propidium iodide. The capsaicin-induced apoptosis was not altered by treatment a VR antagonist, capsazepine. In addition, capsaicin induced a rapid and sustained increase in intracellular Ca^(+2) concentration. This effect of capsaicin was significantly inhibited by BAPTA/AM, an intracellular Ca^(+2) chelator, but not changed by capsazepine. These results suggest that capsaicin induced apoptotic cell dealth through intracellular Ca^(+2) increase in human hepatoma cells, and that VRs may not be involved in the mechanism of the capsaicin-induced apoptosis.
